    Object-Oriented Dynamics Learning through Multi-Level Abstraction

    Object-based approaches for learning action-conditioned dynamics has demonstrated promise for generalization and interpretability. However, existing approaches suffer from structural limitations and optimization difficulties for common environments with multiple dynamic objects. In this paper, we present a novel self-supervised learning framework, called Multi-level Abstraction Object-oriented Predictor (MAOP), which employs a three-level learning architecture that enables efficient object-based dynamics learning from raw visual observations. We also design a spatial-temporal relational reasoning mechanism for MAOP to support instance-level dynamics learning and handle partial observability. Our results show that MAOP significantly outperforms previous methods in terms of sample efficiency and generalization over novel environments for learning environment models. We also demonstrate that learned dynamics models enable efficient planning in unseen environments, comparable to true environment models.     Genetic diversity and phylogenetic analyses of 11 cohorts of captive rhesus macaques from Chinese zoos

    Rhesus macaques are raised in almost every Chinese zoo due to their likeability and ease in feeding; however, little is yet known about the genetic diversity of rhesus macaques in captivity. In this study, a 475-base pair nucleotide sequence of the mitochondrial DNA control region was obtained from the fecal DNA of 210 rhesus macaque individuals in captivity. A total of 69 haplotypes were defined, 51 of which (73.9%) were newly identified. Of all haplotypes, seven were shared between two zoos, and 62 haplotypes (89.8%) appeared only in a specific zoo, indicating a low rate of animal exchange between Chinese zoos. Moreover, there was a relatively high level of genetic diversity among the rhesus macaques (Hd = 0.0623 ± 0.0009, Pi = 0.979 ± 0.003, K = 28.974). Phylogenetic analysis demonstrated that all haplotypes were clearly clustered into two major haplogroups—Clade A (southeastern China) and Clade B (southwestern China)—and each major clade contained several small sub-haplogroups. The haplotypes of rhesus macaques from the same zoo were not clustered together for the most part, but scattered among several subclades on the phylogenetic tree. This indicates that the rhesus macaques in most Chinese zoos may originat from a diverse collection of geographical areas. Our results demonstrate that zoos play an important role in the conservation of the genetic diversity of rhesus macaques, as well as provide useful information on the genetic management of captive rhesus macaques

Dále se věnuji úkolům, které mi byly zadány s jejich implementací a postupem řešení problémů, které se objevily při vývoji. Závěr práce je věnován zhodnocení získaných zkušeností a dosažených výsledků.Purpose of this bachelor thesis is to describe a professional practice in company HS Interactive s.r.o. Practice was focused on the development of mobile application for the operating system Android. The application is a mobile client for social network MatchToMe. In the introduction I describe reasons that led to the selection of professional practice. Then I describe tasks that I have been awarded with their implementations and process of solution issues that have emerged during development.     Productive Hepatitis C Virus Infection of Stem Cell-Derived Hepatocytes Reveals a Critical Transition to Viral Permissiveness during Differentiation

    Primary human hepatocytes isolated from patient biopsies represent the most physiologically relevant cell culture model for hepatitis C virus (HCV) infection, but these primary cells are not readily accessible, display individual variability, and are largely refractory to genetic manipulation. Hepatocyte-like cells differentiated from pluripotent stem cells provide an attractive alternative as they not only overcome these shortcomings but can also provide an unlimited source of noncancer cells for both research and cell therapy. Despite its promise, the permissiveness to HCV infection of differentiated human hepatocyte-like cells (DHHs) has not been explored. Here we report a novel infection model based on DHHs derived from human embryonic (hESCs) and induced pluripotent stem cells (iPSCs). DHHs generated in chemically defined media under feeder-free conditions were subjected to infection by both HCV derived in cell culture (HCVcc) and patient-derived virus (HCVser). Pluripotent stem cells and definitive endoderm were not permissive for HCV infection whereas hepatic progenitor cells were persistently infected and secreted infectious particles into culture medium. Permissiveness to infection was correlated with induction of the liver-specific microRNA-122 and modulation of cellular factors that affect HCV replication. RNA interference directed toward essential cellular cofactors in stem cells resulted in HCV-resistant hepatocyte-like cells after differentiation. The ability to infect cultured cells directly with HCV patient serum, to study defined stages of viral permissiveness, and to produce genetically modified cells with desired phenotypes all have broad significance for host-pathogen interactions and cell therapy

    Genetic Types and Source of the Upper Paleozoic Tight Gas in the Hangjinqi Area, Northern Ordos Basin, China

    The molecular and stable isotopic compositions of the Upper Paleozoic tight gas in the Hangjinqi area in northern Ordos Basin were investigated to study the geochemical characteristics. The tight gas is mainly wet with the dryness coefficient (C1/C1–5) of 0.853–0.951, and δ13C1 and δ2H-C1 values are ranging from -36.2‰ to -32.0‰ and from -199‰ to -174‰, respectively, with generally positive carbon and hydrogen isotopic series. Identification of gas origin indicates that tight gas is mainly coal-type gas, and it has been affected by mixing of oil-type gas in the wells from the Shilijiahan and Gongkahan zones adjacent to the Wulanjilinmiao and Borjianghaizi faults. Gas-source correlation indicates that coal-type gas in the Shiguhao zone displays distal-source accumulation. It was mainly derived from the coal-measure source rocks in the Upper Carboniferous Taiyuan Formation (C3t) and Lower Permian Shanxi Formation (P1s), probably with a minor contribution from P1s coal measures from in situ Shiguhao zone. Natural gas in the Shilijiahan and Gongkahan zones mainly displays near-source accumulation. The coal-type gas component was derived from in situ C3t-P1s source rocks, whereas the oil-type gas component might be derived from the carbonate rocks in the Lower Ordovician Majiagou Formation (O1m)

    A new ovoviviparous rat snake species of the genus Elaphe (Squamata: Colubridae) from western Sichuan, China

    The genus Elaphe is a highly diverse snake group with wide distribution in Eurasia. Here, based on morphological comparisons and molecular data, we describe a new species of this genus from western Sichuan, China. Bayesian inference and maximum-likelihood analyses of two mitochondrial DNA fragments (CO1 and 12S) showed that the new taxon differs from its congeners (CO1-based p-distance ≥ 4.1%). Morphologically and ecologically, the new species can be diagnosed from other species by a combination of the following characters: 1) medium body size, 2) midbody dorsal scales in 23 rows generally, not keeled; 3) ventrals 169–180 and subcaudal pairs 56–63; 4) first preocular docked with frontal; 5) internasal scales approximately trapezoidal; 6) length of gap between internasals more than 3/5 that between prefrontal; 7) length of tip of snout to frontal slightly larger than length of frontal; 8) ovoviviparous. Currently, the new species is known only from the Sichuan and Shaanxi provinces in China. This new species brings the total number of species in the genus Elaphe to 18
